Activities planned in the

first year of the project

 

Growing of the plants and sampling of leaves

(for DNA analyses) in the phase of 4-6 real leaves.

 

 

 

Isolation of DNA according to CTAB method

(Doyle and Doyle, 1987) modified by Grljušić

(2003).

 

 

Checking of the quality of DNA in agarosis gel.

 

 

 

Determination of the quantity of isolated DNA.

 

 

 

 

Presentation of the project at the international

conference.

 

 

 

Preparation of morphological data for statistical

analysis.

 

Realisation of the activities for

the first year of the project

 

440 plants (20 plants per population) were planted

in the controlled environment. Leaves for DNA

analyses were sampled from these plants.

 

DNA was isolated from lyophilized leaves,

according to modified CTAb method.

 

 

The quality of DNA was estimated as high.

 

 

The DNA amount varied among samples.

The concentration of DNA was reduced to be the

same amount of 50 ng/ul in all samples.

 

The research was presented (oral presentation) at

48th national and 8th international symposium of

agronomist in Dubrovnik (17.- 28. February 2013).

 

The data of morphological measurements and

observations were entered into the computer.

Their accuracy was checked. The data were

arranged according to the statistical software

requirements.
